Immunoprecipitation - Anti-IRE1 (phospho S724) antibody [EPR23107-16] - BSA and Azide free (ab267395)
IRE1 (phospho S724) was immunoprecipitated from 0.35 mg HeLa (Human cervix adenocarcinoma epithelial cell) treated with 10mM DTT for 1h whole cell lysate 10µg with ab243665 at 1/30 dilution (2µg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab243665 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366) was used at 1/5000 dilution.

Lane 2: ab243665 IP in HeLa treated with 10mM DTT for 1h whole cell lysate
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab243665 in HeLa treated with 10mM DTT for 1h whole cell lysateBlocking and dilution buffer and concentration: 5% NFDM/TBST.
Exposure time: 3 mins.
Phosphorylation of IRE1 at S724 can be induced by DTT treatment (PMID: 31453810).
