Immunocytochemistry analysis of U-937 (human histiocytic lymphoma monocyte) labeling Liver Carboxylesterase 1/CES1 with purified ab53008 at 1/50 dilution (5 µg/ml). Cells were fixed with 4% Paraformaldehyde and permeabilised with 0.1% tritonX-100. Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) at 1/1000 (2 µg/ml) was used as the secondary antibody. ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1/200 (2.15 µg/ml) was used as counterstain. Nuclei were stained blue with DAPI.
Negative control: PBS instead of the primary antibody. -
Purified ab53008 at 1/100 dilution (2µg) immunoprecipitating Liver Carboxylesterase 1/CES1 in U-937 whole cell lysate.
Lane 1 (input): U-937 (Human histiocytic lymphoma monocyte) whole cell lysate 10µg
Lane 2 (+): ab53008 + U-937 whole cell lysate.
Lane 3 (-): Rabbit monoclonal IgG (ab172730) instead of ab53008 in U-937 whole cell lysate.
VeriBlot for IP Detection Reagent (HRP) (ab131366) (1/1000 dilution) was used for Western blotting.
Blocking Buffer and concentration: 5% NFDM/TBST.
Diluting buffer and concentration: 5% NFDM/TBST.
Observed band size: 60 kDa -
ab53008, at a 1/250 dilution, staining Human SOAT 1 in Liver using Immunohistochemistry, Paraffin embedded tissue.
Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
