Immunocytochemistry/ Immunofluorescence - Anti-Ionotropic Glutamate receptor 2 antibody [EPR5032] (ab133477)ab133477 staining GluR2 in SK-N-SH cells treated with VU 0360223 (ab120803), by ICC/IF. Decrease of GluR2 expression correlates with increased concentration of VU 0360223, as described in literature.
The cells were incubated at 37°C for 6h in media containing different concentrations of ab120803 (VU 0360223) in DMSO, fixed with 4% formaldehyde for 10 minutes at room temperature and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with ab133477 (1/200 dilution was performed overnight at 4°C in PBS containing 1% BSA and 0.1% tween. A DyLight 488 anti-rabbit polyclonal antibody (ab96899) at 1/250 dilution was used as the secondary antibody. Nuclei were counterstained with DAPI and are shown in blue. -
Flow cytometry analysis of SH-SY5Y ( Human neuroblastoma epithelial cell) cells labeling Ionotropic Glutamate receptor 2 (red) with ab133477 at a 1/200 dilution. Cells were fixed with 4% paraformaldehyde and permeabilized with 90% methanol. A goat anti-rabbit IgG (Alexa Fluor® 488) (ab150077) was used as the secondary antibody at a 1/2000 dilution. Black - Rabbit monoclonal IgG (Black) (ab172730). Blue (unlabeled control) - Cell without incubation with primary antibody and secondary antibody (Blue).
