Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.
The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.
One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Immunohistochemical analysis of paraffin-embedded human spleen tissue labeling CD226 with ab212076 at 1/2000 dilution, followed by Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Positive staining of platelets and endothelium in human spleen is observed. Counter stained with hematoxylin.
Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Rabbit specific IHC polymer detection kit HRP/DAB (ab209101).
Perform heat-induced antigen retrieval using ab208572 (Universal HIER antigen retrieval reagent).

All lanes : Anti-CD226 antibody [EPR21172] (ab212076) at 1/1000 dilution
Lane 1 : HEL (human bone marrow erythroleukemia cell line) whole cell lysate
Lane 2 : Human fetal spleen lysate
Lane 3 : Human thymus lysate
Lysates/proteins at 20 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/100000 dilution
Predicted band size: 39 kDa
Observed band size: 60-80 kDa why is the actual band size different from the predicted?Exposure time : Lane 1: 37 seconds; Lane 2: 15 seconds; Lane 3: 3 minutes.
Blocking/Dilution buffer: 5% NFDM/TBST.
The different molecular masses observed in different tissues or cell lines may be due to different level of CD226 glycosylation (PMID: 8673704).

Immunohistochemical analysis of paraffin-embedded human bone marrow tissue labeling CD226 with ab212076 at 1/2000 dilution, followed by Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Cytoplasmic staining in megakaryocytes of human bone marrow (PMID: 15693793) is observed. Counter stained with hematoxylin.
