Mouse Nidogen-1 ELISA Kit (Entactin) (ab270203) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Nidogen-1 (Entactin) protein in cell culture media, cell culture supernatant, edta plasma, serum, cit plasma, and hep plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Nidogen-1 (Entactin) with 44.15 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Nidogen-1 is a glycoprotein found in the basement membrane. Nidogen-1 is a part of a network of proteins within the cell and is thought to be involved in cell-extracellular matrix interactions. Nidogen-1 is a ubiquitous and abundant protein, in contrast to nidogen-2 which has distinct expression patterns. Mice lacking both nidogens die shortly after birth due to lung and heart problems.

The Nidogen-1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Nidogen-1 were measured in duplicates, interpolated from the Nidogen-1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, plasma (citrate) 2.5%, plasma (heparin) 2.5%, and plasma (EDTA) 2.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Nidogen-1 concentration was determined to be 59.56 ng/mL in serum, 165.19 ng/mL in plasma (citrate), 199.81 ng/ml in plasma (heparin), and 167.56 ng/ml in plasma (EDTA).

Undiluted samples are as follows: heart supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Nidogen-1 concentration was determined to be 27.66 ng/mL in heart supernatant.
