Human 14-3-3 tau ELISA Kit (ab246543) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of 37694 tau protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human 37694 tau with 6.1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 14-3-3 Tau, also known as 14-3-3- theta, is a 245- amino acid containing adapter protein encoded by the gene YWHAQ. 14-3-3 tau belongs to the 14-3-3 family of proteins that are involved in a broad range of cell signaling pathways by binding to numerous other proteins, generally by recognition of either a phosphoserine or phosphothreonine motif. 14-3-3 tau is highly conserved across species and has been shown to be involved in cellular processes including metabolism, protein trafficking, signal transduction, and apoptosis.

Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. Negatively regulates the kinase activity of PDPK1. -
Tissue specificity
Abundantly expressed in brain, heart and pancreas, and at lower levels in kidney and placenta. Up-regulated in the lumbar spinal cord from patients with sporadic amyotrophic lateral sclerosis (ALS) compared with controls, with highest levels of expression in individuals with predominant lower motor neuron involvement. -

The 14-3-3 tau standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of 14-3-3 tau were measured in duplicates, interpolated from the 14-3-3 tau standard curves and corrected for sample dilution. Undiluted samples are as follows: A549 cell extract 0.6 µg/mL, Raji cell extract 0.3 µg/mL, and NIH/3T3 cell extract 0.6 µg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
