Function
Tetrodotoxin-resistant channel that mediates the voltage-dependent sodium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a sodium-selective channel through which sodium ions may pass in accordance with their electrochemical gradient. Plays a role in neuropathic pain mechanisms. -
Tissue specificity
Expressed in the dorsal root ganglia and sciatic nerve. -
Involvement in disease
Episodic pain syndrome, familial, 2 -
Sequence similarities
Belongs to the sodium channel (TC 1.A.1.10) family. Nav1.8/SCN10A subfamily.
Contains 1 IQ domain. -
Domain
The sequence contains 4 internal repeats, each with 5 hydrophobic segments (S1,S2,S3,S5,S6) and one positively charged segment (S4). Segments S4 are probably the voltage-sensors and are characterized by a series of positively charged amino acids at every third position. -
Post-translational
modificationsUbiquitinated by NEDD4L; which promotes its endocytosis.
Phosphorylation at Ser-1451 by PKC in a highly conserved cytoplasmic loop slows inactivation of the sodium channel and reduces peak sodium currents. -
Cellular localization
Cell membrane. It can be translocated to the cell membrane through association with S100A10. - Information by UniProt

All lanes : Anti-Nav1.8/SCN10A antibody (ab139510) at 1 µg/ml (Milk blocking - 3%)
Lane 1 : Brain (Mouse) Tissue Lysate
Lane 2 : Cerebellum Mouse Tissue Lysate
Lane 3 : Cerebellum Rat Tissue Lysate
Lysates/proteins at 20 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/10000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 221 kDa
Observed band size: 260 kDa why is the actual band size different from the predicted?
Exposure time: 20 minutesNav1.8 contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted.
This blot was produced using a 3-8% Tris Acetate gel under the TA buffer system. The gel was run at 150V for 60 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% Milk before being incubated with ab139510 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
