General notes
Reproducibility is key to advancing scientific discovery and accelerating scientists’ next breakthrough.
Abcam is leading the way with our range of recombinant antibodies, knockout-validated antibodies and knockout cell lines, all of which support improved reproducibility.
We are also planning to innovate the way in which we present recommended applications and species on our product datasheets, so that only applications & species that have been tested in our own labs, our suppliers or by selected trusted collaborators are covered by our Abpromise™ guarantee.
In preparation for this, we have started to update the applications & species that this product is Abpromise guaranteed for.
We are also updating the applications & species that this product has been “predicted to work with,” however this information is not covered by our Abpromise guarantee.
Applications & species from publications and Abreviews that have not been tested in our own labs or in those of our suppliers are not covered by the Abpromise guarantee.
Please check that this product meets your needs before purchasing. Our Abpromise guarantee covers the use of ab138334 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Application Abreviews Notes WB Use a concentration of 1 µg/ml. Detects a band of approximately 75 kDa (predicted molecular weight: 65 kDa). Target
-
Function
Putative phospholipase. -
Tissue specificity
Ubiquitously expressed, with highest levels in heart, brain and liver. -
Sequence similarities
Belongs to the phospholipase B-like family. -
Post-translational
modificationsThe p76 protein is synthesized as a 80 kDa precursor which is then processed into a N-terminal 32 kDa form and a C-terminal 45 kDa form.
Glycosylated; contains mannose 6-phosphate sugars. -
Cellular localization
Lysosome lumen. - Information by UniProt

All lanes : Anti-PLBD2 antibody (ab138334) at 1 µg/ml
Lane 1 : Spleen (Rat) Tissue Lysate
Lane 2 : Lung (Rat) Tissue Lysate
Lysates/proteins at 10 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/10000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 65 kDa
Observed band size: 75 kDa why is the actual band size different from the predicted?
Additional bands at: 15 kDa (possible cleavage fragment), 40 kDa (possible cleavage fragment)
Exposure time: 6 minutesThe band observed at 40 kDa could potentially be a cleaved form of PLBD2, representing the 346 amino acid chain called "Putative phospholipase B-like 2 40 kDa form".
The band observed at 15 kDa could potentially be a cleaved form of PLBD2, representing the 81 amino acid chain called "Putative phospholipase B-like 2 15 kDa form".
This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab138334 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
