Specificity
The specificity of the NPY antiserum was determined by radioimmunoassay. Synthetic peptides, somatostatin-14, leu-enkephalin, met-enkephalin, oxytocin, Arg8-vasopressin, vasoactive intestinal peptide, angiotensin II demonstrated less than 0.005% cross- reactivity and substance P less than 0.02%. Peptide YY and bovine pancreatic polypeptide demonstrated approx. 36% and 0.05% cross- reactivity respectively. Human peptide YY share 65% homology amino acid sequence for human NPY. Synthetic NPY completely abolished radio labelled NPY binding to NPY antibody in RIA and antibody binding to tissue antigen. -
