Rat Myoglobin ELISA Kit (ab260068) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Myoglobin protein in edta plasma, serum, cell culture media, tissue extracts, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat Myoglobin with 0.1 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Myoglobin, is a 18.4 kDa heme protein, found in both cardiac and skeletal muscle that functions as a reserve supply of oxygen by facilitating the movement of oxygen within muscles. Damage to either type of muscle following conditions such as trauma, ischemia, and diseases that cause myopathy, is associated with the release of myoglobin into serum. Specifically, following cardiac necrosis associated with myocardial infarction (MI), myoglobin is one of the first markers to rise above normal levels. Myoglobin levels increase measurably above baseline within 2-4 hours post-infarct, peaking at 9-12 hours, and returning to baseline within 24-36 hours. Rat Myoglobin shares 83%, 93%, 77%, and 84% sequence homology with human, mouse, bovine, and monkey respectively.

The Myoglobin standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Myoglobin were measured in duplicates, interpolated from the Myoglobin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 100% and plasma (EDTA) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Myoglobin concentration was determined to be 10.60 ng/mL in serum and 16.61 ng/mL in plasma (EDTA).

The concentrations of Myoglobin were measured in duplicates, interpolated from the Myoglobin standard curves and corrected for sample dilution. Undiluted samples are as follows: heart tissue culture supernatant 1%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Myoglobin concentration was determined to be 1927.15 ng/mL in heart tissue culture supernatant.

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Myoglobin concentration was determined to be 5.28 ng/mL in heart extract and 5.87 ng/mL in skeletal muscle extract.
