Product overview
Mouse COX2 ELISA Kit (ab210574) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of COX2 protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse COX2 with 97 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
Cyclooxygenase-2 (COX2) also known as prostaglandin G/H synthase 2 (PGHS2) is a 70 kDa microsomal enzyme that belongs to the prostaglandin G/H synthase family. It is inducibly-expressed by a number of cell types, including fibroblasts, vascular smooth muscle cells, endothelium, and monocytes. Functionally, COX2 is a homodimer that catalyzes two steps in the conversion of arachadonic acid to prostaglandin H2. Mature mouse COX2 is 587 amino acids (aa) in length and contains one EGF-like domain (aa 18-55).

Function
Mediates the formation of prostaglandins from arachidonate. May have a role as a major mediator of inflammation and/or a role for prostanoid signaling in activity-dependent plasticity. -
Pathway
Lipid metabolism; prostaglandin biosynthesis. -
Sequence similarities
Belongs to the prostaglandin G/H synthase family.
Contains 1 EGF-like domain. -
Post-translational
modificationsS-nitrosylation by NOS2 (iNOS) activates enzme activity. S-nitrosylation may take place on different Cys residues in addition to Cys-561. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of mouse COX2 were measured in duplicate and interpolated from the COX2 standard curve then corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean COX2 concentration was determined to be 0.203 ng/mL in unstimulated extract and 5.3 ng/mL in LPS stimulated Raw 264.7 extract.
