Product overview
ab126448 is a very rapid, convenient and sensitive assay kit that can monitor the activation or function of important biological pathways in cell lysates. By determining phosphorylated Mek1 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of phospho-Mek1 (Ser217/221) and total Mek1 in human, mouse and rat cell lysates (help normalize the results of phospho-Mek1 from different cell lysate being compared). An anti-pan Mek1 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and Mek1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and anti-Mek1 (Ser217/221) or anti-pan-Mek1 antibody is used to detect phosphorylated Mek1 (Ser217/221) or total Mek1. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Mek1 (Ser217/221) or total Mek1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Involvement in disease
Defects in MAP2K1 are a cause of cardiofaciocutaneous syndrome (CFC syndrome) [MIM:115150]; also known as cardio-facio-cutaneous syndrome. CFC syndrome is characterized by a distinctive facial appearance, heart defects and mental retardation. Heart defects include pulmonic stenosis, atrial septal defects and hypertrophic cardiomyopathy. Some affected individuals present with ectodermal abnormalities such as sparse, friable hair, hyperkeratotic skin lesions and a generalized ichthyosis-like condition. Typical facial features are similar to Noonan syndrome. They include high forehead with bitemporal constriction, hypoplastic supraorbital ridges, downslanting palpebral fissures, a depressed nasal bridge, and posteriorly angulated ears with prominent helices. The inheritance of CFC syndrome is autosomal dominant. -
