Human SMAC ELISA Kit (ab246533) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SMAC protein in cell culture extracts and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human SMAC with 5.1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. - Single-wash protocol reduces assay time to 90 minutes or less

Promotes apoptosis by activating caspases in the cytochrome c/Apaf-1/caspase-9 pathway. Acts by opposing the inhibitory activity of inhibitor of apoptosis proteins (IAP). -
Tissue specificity
Ubiquitously expressed with highest expression in testis. Expression is also high in heart, liver, kidney, spleen, prostate and ovary. Low in brain, lung, thymus and peripheral blood leukocytes. -

The SMAC standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Interpolated concentrations of native SMAC in human Jurkat cell extract and testis tissue extract based on a 25 µg/mL extract load.The concentrations of SMAC were measured in duplicate and interpolated from the SMAC standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SMAC concentration was determined to be 1,009 pg/mL in Jurkat cell extract and 868 pg/mL in human testis tissue extract.
