Human M-CSF ELISA Kit (ab245714) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of M-CSF protein in hep plasma, saliva, serum, urine, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human M-CSF with 3.8 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Granulocyte/macrophage colony-stimulating factors are cytokines that act in hematopoiesis by controlling the production, differentiation, and function of 2 related white cell populations of the blood, the granulocytes and the monocytes-macrophages. CSF-1 induces cells of the monocyte/macrophage lineage. It plays a role in immunological defenses, bone metabolism, lipoproteins clearance, fertility and pregnancy. -

The M-CSF standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of M-CSF were measured in duplicates, interpolated from the M-CSF standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 2%, plasma (heparin) 2.5%, and PBMC supernatant 7.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean M-CSF concentration was determined to be 11.5 ng/mL in serum, 6.7 ng/mL in plasma (heparin), and 2.4 ng/mL in stimulated PBMC supernatant.

Undiluted samples are as follows: saliva 25%, urine 1:200, and A549 cell supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean M-CSF concentration was determined to be 0.9 ng/mL in saliva, 28.1 ng/mL in urine, and 0.89 ng/mL in A549 cell supernatant.
