Product overview
Human LKB1 ELISA Kit (ab233612) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of LKB1 protein in cell culture extracts and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human LKB1 with 1 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Research areas
-
Function
Essential role in G1 cell cycle arrest. Phosphorylates and activates members of the AMPK-related subfamily of protein kinases. Tumor suppressor. -
Tissue specificity
Ubiquitously expressed. Strongest expression in testis and fetal liver. -
Involvement in disease
Defects in STK11 are a cause of Peutz-Jeghers syndrome (PJS) [MIM:175200]. PJS is a rare hereditary disease in which there is predisposition to benign and malignant tumors of many organ systems. PJS is an autosomal dominant disorder characterized by melanocytic macules of the lips, multiple gastrointestinal hamartomatous polyps and an increased risk for various neoplasms, including gastrointestinal cancer.
Defects in STK11 have been associated with testicular tumors (TEST) [MIM:273300]. A common solid malignancy in males. Germ cell tumors of the testis constitute 95% of all testicular neoplasms. -
Sequence similarities
Belongs to the protein kinase superfamily. CAMK Ser/Thr protein kinase family. LKB1 subfamily.
Contains 1 protein kinase domain. -
Post-translational
modificationsPhosphorylated by a cAMP-dependent protein kinase. -
Cellular localization
Nucleus. Cytoplasm. Relocates to the cytoplasm when bound to CAB39 and STRAD or CAB39 and ALS2CR2. - Information by UniProt

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The LKB1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
Interpolated concentrations of native LKB1 in human MCF-7 cell extract based on a 3,000 µg/mL extract load and HEK293T cell extract based on a 1,500 µg/mL extract load.
The concentrations of LKB1 were measured in duplicate and interpolated from the LKB1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LKB1 concentration was determined to be 27 ng/mL in MCF-7 cell extract and 46.3 ng/mL in HEK293T cell extract.
-
Interpolated concentrations of native LKB1 in mouse RAW264.7 and rat PC-12 cell extract, extract based on a 750 µg/mL extract load.
The concentrations of LKB1 were measured in duplicate and interpolated from the LKB1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LKB1 concentration was determined to be 61 ng/mL in mouse RAW 264.7 cell extract and 11 ng/mL in rat PC12.
