Function
Isoform Alpha is a retrovirus restriction factor, which mediates species-specific, early block to retrovirus infection. Targets retroviral capsid soon after entry into the cell, and prevents reverse transcription of the virus RNA genome. Isoform Alpha trimers may make multiple contacts with the hexameric lattice of CA proteins which constitute the surface of retrovirion core, and somehow inactivate the virus. Restricts efficiently infection by N-MLV, but not HIV-1. May have E3 ubiquitin-protein ligase activity. -
Pathway
Protein modification; protein ubiquitination. -
Sequence similarities
Belongs to the TRIM/RBCC family.
Contains 1 B box-type zinc finger.
Contains 1 B30.2/SPRY domain.
Contains 1 RING-type zinc finger. -
Domain
The RING-type zinc finger domain mediates binding to an E2 ubiquitin-conjugating enzyme. -
Post-translational
modificationsUbiquitinates itself in a RING finger- and UBE2D2-dependent manner (in vitro). -
Cellular localization
Cytoplasm > P-body. Cytoplasmic bodies. - Information by UniProt

All lanes : Anti-TRIM5 alpha antibody (ab109709) at 1 µg/ml
Lane 1 : Human placenta tissue lysate - total protein (ab29745)
Lane 2 : Tonsil (Human) Tissue Lysate
Lane 3 : Human thymus tissue lysate - total protein (ab30146)
Lysates/proteins at 10 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) preadsorbed (ab97080) at 1/5000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 56 kDa
Observed band size: 56 kDa
Additional bands at: 31 kDa (possible isoform), 46 kDa (possible isoform)
Exposure time: 2 minutes
This antibody was raised against an immunogen that is predicted to recognize isoforms Alpha, Beta and Epsilon of TRIM5. The predicted molecular weights of isoforms Alpha, Beta and Epsilon are 56, 46 and 31 kDa respectively. -
IHC image of TRIM5 alpha staining in human tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab109709, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
