Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.
The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.
One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Flow cytometric analysis of human peripheral blood mononuclear cells (PBMCs) labeling TIM 3 with ab210543 at 1/500 dilution (right panel) compared with a Rabbit IgG, monoclonal [EPR25A] - Isotype Control (ab172730) (left panel). Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) at 1/2000 dilution was used as the secondary antibody.
Co-staining with anti-CD14 (y-axis). CD14+ PBMCs were positive for TIM 3 (PMID 22426209). Gated on total viable cells.

Anti-TIM 3 antibody [EPR20767] (ab210543) at 1/1000 dilution + Fc-tagged human TIM 3 recombinant protein (aa22-200) at 0.01 µg
Secondary
Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/100000 dilution
Developed using the ECL technique.
Predicted band size: 33 kDa
Observed band size: 75 kDa why is the actual band size different from the predicted?
Exposure time: 3 secondsBlocking/Dilution buffer: 5% NFDM/TBST.

Anti-TIM 3 antibody [EPR20767] (ab210543) at 1/1000 dilution + Daudi (human Burkitt's lymphoma cell line) whole cell lysate at 10 µg
Secondary
Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/100000 dilution
Developed using the ECL technique.
Predicted band size: 33 kDa
Observed band size: 50 kDa why is the actual band size different from the predicted?
Exposure time: 15 secondsBlocking/Dilution buffer: 5% NFDM/TBST.
Strong signal detected in Daudi cells after 15s exposure. Please note that under our experimental conditions (overexposure >3min), a faint band at ~70kDa was detected in Jurkat T cells (previously reported as a TIM 3 negative cell line PMID: 21807895).

TIM 3 was immunoprecipitated from 0.35 mg of Daudi (human Burkitt's lymphoma cell line) whole cell lysate with ab210543 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab210543 at 1/500 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/1000 dilution.
Lane 1: Daudi whole cell lysate 10 µg (Input).
Lane 2: ab210543 IP in Daudi whole cell lysate.
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab210543 in Daudi whole cell lysate.Exposure time: 10 seconds.
Blocking and dilution buffer and concentration: 5% NFDM/TBST.
