Lane 1: Wild-type HAP1 cell lysate (20 µg)
Lane 2: LRP6 knockout HAP1 cell lysate (20 µg)
Lane 3: HeLa (human epithelial cell line from cervix adenocarcinoma) cell lysate (20 µg)
Lane 4: HepG2 (human liver hepatocellular carcinoma cell line) cell lysate (20 µg)
Lanes 1 - 4: Merged signal (red and green). Green - ab134146 observed at 220 kDa. Red - loading control, ab18058, observed at 124 kDa.
ab134146 was shown to recognize LRP6 when LRP6 knockout samples were used, along with additional cross-reactive bands. Wild-type and LRP6 knockout samples were subjected to SDS-PAGE. ab134146 and ab18058 (loading control to Vinculin) were diluted 1/10000 and 1/1000 respectively, and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging. This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab134146).
