IRF7 was immunoprecipitated from 0.35 mg of Jurkat (treated with 100 nM IFN alpha for 18 hours, then added 10 μM MG-132 for another 6 hours) whole cell lysate with ab238137 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab238137 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366), was used as secondary antibody at 1/5000 dilution.
Lane 1: Jurkat (treated as above) whole cell lysate 10 μg (Input).
Lane 2: ab238137 IP in Jurkat (treated as above) whole cell lysate.
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab238137 in Jurkat (treated as above) whole cell lysate.Blocking and dilution buffer and concentration: 5% NFDM/TBST.
Exposure time: 84 seconds.65-kDa full length IRF7 and 45-kDa C-terminal cleaved IRF7 are observed (PMID: 23175366).
This data was developed using the same antibody clone in a different buffer formulation containing PBS, glycerol, BSA and sodium azide (ab238137).
