Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-GALC antibody [EPR23598-126] (ab240638)
Immunohistochemical analysis of paraffin-embedded Rat cerebrum tissue labeling GALC with ab240638 at 1/2000 dilution followed by a ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Positive staining on rat cerebrum (PMID: 15987783). The section was incubated with ab240638 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.
Secondary antibody only control: Secondary antibody is a ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101).
Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.

Observed band size: 140,50,70,80 kDa why is the actual band size different from the predicted?Blocking and diluting buffer and concentration: 5% NFDM/TBST.
The precursor form of GALC (80 KD) is processed in lysosomes into N-terminal fragment (70 and 50 KD) and C-terminal fragment (30 KD), the observed band at 140 KD is the aggregate GALC, which is consistent to the literature(PMID: 7852280;PMID: 9760019;PMID: 20410102).
Exposure time: 37 seconds.
