Cripto1/CRIPTO was immunoprecipitated from 0.35 mg of NCCIT (human pluripotent embryonic carcinoma cell line) whole cell lysate with ab229016 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab229016 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/5000 dilution.
Lane 1: NCCIT whole cell lysate 10 µg (Input).
Lane 2: ab229016 IP in NCCIT whole cell lysate.
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab229016 in NCCIT whole cell lysate.
Blocking and dilution buffer and concentration: 5% NFDM/TBST.
Exposure time: 2 minutes.

Predicted band size: 21 kDaBlocking/Dilution buffer: 5% NFDM/TBST.
The double bands observed are consistent with what has been described in the literature. The lower band represents the immature form of Cripto1/CRIPTO. The upper band is likely to be the processed glycosylated form (PMID: 27881714 and 20568225).
