Relevance
COQ10 (Coenzyme Q10) is found in a wide variety of foods and is synthesized in all tissues. The de novo biosynthesis of COQ10, from its precursor tyrosine, is a multistage process that requires at least eight vitamins and several trace elements. More complex enzymes have an absolute dependancy on 'cofactors', a large group of molecules to which the coenzymes belong, for their catalyitic function. Coenzyme Q10 is the cofactor for, but not limited to, several mitochondrial enzymes that are central to the supply of adenosine triphosphate (ATP). In addition, COQ10 in its reduced form is a potent antioxidant, and in this capacity, a reduction in COQ10 levels are associtated with a variety of degenerative process including skin aging and neurodegenerative conditions such as Alzhiemer's, Huntingdon's and Parkinson's diseases. -

COQ10B has a predicted molecular weight of 27 kDa; however as this is a novel protein, it may contain uncharacterised modification sites that affect the migration of the protein. IHC image of ab41997 staining COQ10B in Human liver formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab41997, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times. -
ICC/IF image of ab41997 stained HeLa cells. The cells were stained with Mitotracker Orange CMTMRos at a concentration of 500nM in PBS and incubated for 45 minutes at 37°C. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab41997, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Dylight® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
