Function
Component C2 which is part of the classical pathway of the complement system is cleaved by activated factor C1 into two fragments: C2b and C2a. C2a, a serine protease, then combines with complement factor 4b to generate the C3 or C5 convertase. -
Involvement in disease
Defects in C2 are the cause of complement component 2 deficiency (C2D) [MIM:217000]. A deficiency of the complement classical pathway associated with the development of autoimmune disorders, mainly systemic lupus erythematosus. Skin and joint manifestations are common and renal disease is relatively rare. Patients with complement component 2 deficiency are also reported to have recurrent or invasive infections. -

Observed band size: 105 kDa why is the actual band size different from the predicted?
Additional bands at: 28 kDa, 55 kDa. We are unsure as to the identity of these extra bands.
Exposure time: 20 minutes
C2 contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted. Furthermore, the 105 kDa band observed is comparable to the molecular weight seen with other commercially available antibodies this target. -
IHC image of Ab77498 staining in Human Liver Carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with Ab77498, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
