  • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Target

  • Relevance

The budding yeast INO80 complex is a conserved ATP-dependent nucleosome remodeler containing actin-related proteins Arp5 and Arp8. Strains lacking INO80, ARP5, or ARP8 have defects in transcription. Ino80, Arp5, and Arp8 are recruited to an HO-induced DSB, where a phosphorylated form of H2A accumulates. Recruitment of Ino80 is compromised in cells lacking the H2A phosphoacceptor S129. INO80-mediated chromatin remodeling seems to occur directly at DSBs, where it appears to facilitate processing of the lesion.

      Predicted band size: 87 kDa
Observed band size: 100 kDa why is the actual band size different from the predicted?

Using ab12099, bands were present in all S. cerevisiae strains tested except for an arp5 knockout strain. This provides conclusive evidence that the ab12099 recognises Arp5.
