General notes
The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.
One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Function
ATP dependent phosphorylation of adenosine and other related nucleoside analogs to monophosphate derivatives. Serves as a potential regulator of concentrations of extracellular adenosine and intracellular adenine nucleotides. -
Tissue specificity
Widely expressed. Highest level in placenta, liver, muscle and kidney. -
Pathway
Purine metabolism; AMP biosynthesis via salvage pathway; AMP from adenosine 3',5'-bisphosphate: step 1/1. -

All lanes : Anti-ADK antibody (ab88903) at 1/1000 dilution
Lane 1 : Wild-type HAP1 whole cell lysate at 40 µg
Lane 2 : ADK knockout HAP1 whole cell lysate at 40 µg
Lane 3 : HepG2 whole cell lysate at 20 µg
Predicted band size: 41 kDaLanes 1 - 3: Merged signal (red and green). Green - ab88903 observed at 41 kDa. Red - loading control, ab130007, observed at 130 kDa.
ab88903 was shown to recognize ADK in wild-type HAP1 cells as signal was lost at the expected MW in ADK knockout cells. Additional cross-reactive bands were observed in the wild-type and knockout cells. Wild-type and ADK knockout samples were subjected to SDS-PAGE. Ab88903 and ab130007 (Mouse anti-Vinculin loading control) were incubated overnight at 4°C at 1/1000 dilution and 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

All lanes : Anti-ADK antibody (ab88903) at 1 µg/ml
Lane 1 : RAW 264.7 (Mouse leukaemic monocyte macrophage cell line) Whole Cell Lysate
Lane 2 : Human placenta tissue lysate - total protein (ab29745)
Lane 3 : Human liver tissue lysate - total protein (ab29889)
Lane 4 : HepG2 (Human hepatocellular liver carcinoma cell line) Whole Cell Lysate
Lysates/proteins at 10 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) preadsorbed (ab97080) at 1/5000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 41 kDa
Observed band size: 39,41 kDa why is the actual band size different from the predicted?
Exposure time: 8 minutes
Ab88903 is raised against an immunogen that is present in both isoforms of ADK. We hypothesise that the bands observed correspond to the long and short isoforms, which are 41- and 39-kDa respectively.
