ab225486 staining LAG-3 in 293T cells transfected with LAG-3 with GFP tag. The cells were fixed with 4% formaldehyde (10 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab225486 at 1/100 dilution (pseudocolored in white) and ab195889, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 594), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue) and GFP is shown in green.
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Flow cytometric analysis of HEK-293T (Human epithelial cell line from embryonic kidney) cells transfected with a GFP-tagged human LAG3 construct labelling LAG-3 with ab225486 at 1/500 dilution (right) compared with Rabbit IgG (monoclonal) Alexa Fluor® 647 ab199093 (left).
Note: Fresh cells without fixation and permeabilization were used to perform FC testing. Only GFP positive population results in LAG3 positive staining (Q2, right panel).
