Rat uPA ELISA kit (total uPA antigen) (ab272781) is intended for the quantitative determination of total urokinase plasminogen activator in rat plasma, serum, urine, cell culture media, or tissue extracts.
Rat uPA will bind to the capture antibody coated on the microtiter plate. Free and complexed enzyme will react with the capture antibody on the plate. After appropriate washing steps, polyclonal anti-rat uPA primary antibody binds to the captured enzyme. Excess antibody is washed away and bound polyclonal antibody is then reacted with the secondary antibody conjugated to horseradish peroxidase. TMB substrate is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of uPA.

Urokinase type plasminogen activator (uPA) is secreted from cells as a precursor which is activated to the two chain form consisting of an A and B chain. The active high MW form is further processed by removal of an amino terminal fragment to an active low MW form (35 kDa). uPA is a serine protease that activates plasminogen toplasmin. High levels of uPA and plasminogen activator inhibitor type 1 (PAI 1) in breast cancer tissue extracts have been associated with rapid disease progression. The malignant phenotype of prostatic tumor cells correlates with the expression of both uPA and its cell membrane receptor (uPAR). -
