Rat IL-10 ELISA Kit (ab214566) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IL-10 protein in cell culture supernatant, cit plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat IL-10 with 4.89 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The concentrations of IL-10 were measured in duplicates, interpolated from the IL-10 standard curves and corrected for sample dilution. Undiluted samples are as follows: rat PBMC stimulated with PHA (10 µg/mL, 48 hours) supernatant 50%, unstimulated PBMC supernatant 100%, and spleen stimulated with concanavalin A (5 µg/mL, 48 hours) supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-10 concentration was determined to be 1,174 pg/mL in PBMC stimulated with PHA supernatant, 230 pg/mL in unstimulated PBMC supernatant and 789 pg/mL in spleen stimulated with concanavalin A supernatant.

Conditioned media was harvested after 48 hours. IL-10 was measured in 50% unstimulated and PHA stimulated PBMC supernatant. The concentrations of IL-10 were measured in duplicate, interpolated from the IL-1beta standard curves and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-10 concentration was determined to be 1,068 pg/mL in PHA stimulated PBMC supernatant and 227 pg/mL in unstimulated PBMC supernatant.
