Human Progranulin ELISA Kit (ab252364) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Progranulin protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, milk, saliva, serum, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Progranulin with 72.68 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Function
Granulins have possible cytokine-like activity. They may play a role in inflammation, wound repair, and tissue remodeling.
Granulin-4 promotes proliferation of the epithelial cell line A431 in culture while granulin-3 acts as an antagonist to granulin-4, inhibiting the growth. -
Tissue specificity
In myelogenous leukemic cell lines of promonocytic, promyelocytic, and proerythroid lineage, in fibroblasts, and very strongly in epithelial cell lines. Present in inflammatory cells and bone marrow. Highest levels in kidney. -

Defects in GRN are the cause of ubiquitin-positive frontotemporal dementia (UP-FTD) [MIM:607485]; also known as tau-negative frontotemporal dementia linked to chromosome 17. Frontotemporal dementia (FTD) is the second most common cause of dementia in people under the age of 65 years. It is an autosomal dominant neurodegenerative disease. -

The Progranulin standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Progranulin were measured in duplicates, interpolated from the Progranulin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, plasma (citrate) 6.25%, plasma (heparin) 6.25%, and plasma (EDTA) 6.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Progranulin concentration was determined to be 60.9 ng/mL in serum, 47.8 ng/mL in plasma (citrate), 52.2 ng/ml in plasma (heparin), and 49.8 ng/ml in plasma (EDTA).

Interpolated concentrations of native Progranulin in human milk, saliva, and cell culture supernatant samples.
The concentrations of Progranulin were measured in duplicates, interpolated from the Progranulin standard curves and corrected for sample dilution. Undiluted samples are as follows: milk 1: 160, saliva 5%, and OVCAR-3 supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Progranulin concentration was determined to be 360.2 ng/ml in milk, 60.4 ng/ml in saliva, and 24.4 ng/ml in OVCAR-3 supernatant.
