Human Nectin-4 ELISA Kit (ab264607) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Nectin-4 protein in cell lysate, hep plasma, serum, urine, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Nectin-4 with 98.62 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Nectin-4 is a cell adhesion molecule located in the epithelial adherens junctions. Mutations in Nectin-4 lead to Ectodermal dysplasia-syndactyly syndrome, a condition where there is abnormal development of two or more ectodermal structures (like hair, tooth and nails). A fragment of Nectin-4 is shed and circulates in serum in certain types of cancers. Additionally, Nectin-4 is also a receptor for the measles virus.

Function
Seems to be involved in cell adhesion through trans-homophilic and -heterophilic interactions, the latter including specifically interactions with PVRL2/nectin-1. Does not act as receptor for alpha-herpesvirus entry into cells. -
Tissue specificity
Predominantly expressed in placenta. Not detected in normal breast epithelium but expressed in breast carcinoma. -
Involvement in disease
Defects in PVRL4 are the cause of ectodermal dysplasia-syndactyly syndrome type 1 (EDSS1) [MIM:613573]. EDSS1 is a form of ectodermal dysplasia, a heterogeneous group of disorders due to abnormal development of two or more ectodermal structures. EDSS1 is characterized by the association of hair and teeth abnormalities with cutaneous syndactyly of the hands and/or feet. Hair morphologic abnormalities include twists at irregular intervals (pilli torti) and swelling along the shafts, particularly associated with areas of breakage. Dental findings consist of abnormally widely spaced teeth, with peg-shaped and conical crowns. Patients have normal sweating. -

The Nectin-4 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Nectin-4 were measured in duplicates, interpolated from the Nectin-4 standard curves and corrected for sample dilution. Undiluted samples are as follows: urine 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Nectin-4 concentration was determined to be 7.34 ng/mL in urine.
