Human Glycogen Synthase 1 ELISA Kit (ab214031) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Glycogen Synthase 1 protein in tissue extracts, plasma, serum, and cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Glycogen Synthase 1 with 14 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Glycogen Synthase 1 (also known as Glycogen [starch] synthase, muscle) is a glycosyltransferase enzyme involved in converting glucose to glycogen. Mutations in Glycogen Synthase 1 are associated with glycogen storage disease. Mouse and rat Glycogen Synthase 1 have 97% protein sequence identity to human Glycogen Synthase 1.

Involvement in disease
Defects in GYS1 are the cause of muscle glycogen storage disease type 0 (GSD0b) [MIM:611556]; also known as muscle glycogen synthase deficiency. GSD0b is a metabolic disorder characterized by fasting hypoglycemia presenting in infancy or early childhood. The role of muscle glycogen is to provide critical energy during bursts of activity and sustained muscle work. -

The concentrations of Glycogen Synthase 1 were measured in duplicate and interpolated from the Glycogen Synthase 1 standard curve and corrected for sample dilution and extract load. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Glycogen Synthase 1 concentration was determined to be 38 pg/µg HELA extract, 129 pg/µg HEK293 extract and 5.9 pg/µg Liver extract.
