Human FOXO3A ELISA Kit (ab277471) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human FOXO3A in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for Human FOXO3A coated on a 96-well plate. Standards and samples are pipetted into the wells and Human FOXO3A present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Human FOXO3A antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Human FOXO3A bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Post-translational
modificationsIn the presence of survival factors such as IGF-1, phosphorylated on Thr-32 and Ser-253 by AKT1/PKB. This phosphorylated form then interacts with 14-3-3 proteins and is retained in the cytoplasm. Survival factor withdrawal induces dephosphorylation and promotes translocation to the nucleus where the dephosphorylated protein induces transcription of target genes and triggers apoptosis. Although AKT1/PKB doesn't appear to phosphorylate Ser-315 directly, it may activate other kinases that trigger phosphorylation at this residue. Phosphorylated by STK4 on Ser-209 upon oxidative stress, which leads to dissociation from YWHAB/14-3-3-beta and nuclear translocation. Phosphorylated by PIM1. -
