Human ERCC1 ELISA Kit (ab210582) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of ERCC1 protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human ERCC1 with 58 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
ERCC1 is a DNA repair endonuclease. It plays a role in the first stage of the repair of interstrand cross-links by 5’-incision. ERCC1 is also essential for homology-directed repair of DNA double strand breaks.

Involvement in disease
Defects in ERCC1 are the cause of cerebro-oculo-facio-skeletal syndrome type 4 (COFS4) [MIM:610758]. COFS is a degenerative autosomal recessive disorder of prenatal onset affecting the brain, eye and spinal cord. After birth, it leads to brain atrophy, hypoplasia of the corpus callosum, hypotonia, cataracts, microcornea, optic atrophy, progressive joint contractures and growth failure. Facial dysmorphism is a constant feature. Abnormalities of the skull, eyes, limbs, heart and kidney also occur. -

The concentrations of ERCC1 were measured in duplicate and interpolated from the ERCC1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ERCC1 concentration was determined to be 1,146 pg/mL in HepG2 extracts and 490 pg/mL in HUVEC extracts.
