Product overview
Human BACE-1 ELISA Kit is designed for the quantitative determination of BACE-1 in cell culture supernatants, plasma and serum samples.
ΔNote: Human BACE-1 concentration is low in normal serum/plasma and may not be detectable in this assay.
This assay employs an antibody specific for human BACE-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and BACE-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human BACE-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of BACE-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
This ELISA antibody pair detects human BACE-1, it also can recognize mouse BACE-1.

Function
Responsible for the proteolytic processing of the amyloid precursor protein (APP). Cleaves at the N-terminus of the A-beta peptide sequence, between residues 671 and 672 of APP, leads to the generation and extracellular release of beta-cleaved soluble APP, and a corresponding cell-associated C-terminal fragment which is later released by gamma-secretase. -

Expressed at high levels in the brain and pancreas. In the brain, expression is highest in the substantia nigra, locus coruleus and medulla oblongata. -
