Product overview
Human AKR1B10 ELISA Kit (ab254506) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of AKR1B10 protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human AKR1B10 with 10.3 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
Aldo-Keto Reductase Family-1 member B10 (AKR1B10), also referred to as Aldose Reductase-like protein, is an 316- amino acid enzyme encoded by the AKR1B10 gene. AKR1B10 belongs to the aldo/keto reductase superfamily, where AKR1B10’s function is to catalyze the NADPH-dependent reduction of carbonyls to the corresponding alcohols. Found to be highly expressed in the small intestine and colon, AKR1B10 is thought to be responsible in detoxifying dietary and lipid derived carbonyls and may play a role in liver carcinogenesis. AKR1B10 has no counterpart in mouse or rat species.

Function
Acts as all-trans-retinaldehyde reductase. Can efficiently reduce aliphatic and aromatic aldehydes, and is less active on hexoses (in vitro). May be responsible for detoxification of reactive aldehydes in the digested food before the nutrients are passed on to other organs. -
Tissue specificity
Found in many tissues. Highly expressed in small intestine, colon and adrenal gland. -
Sequence similarities
Belongs to the aldo/keto reductase family. -
Cellular localization
Lysosome. Secreted. Secreted through a lysosome-mediated non-classical pathway. - Information by UniProt

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The AKR1B10 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
Interpolated concentrations of native AKR1B10 in human A549 cell extract based on a 0.31 µg/mL extract load. The concentrations of AKR1B10 were measured in duplicate and interpolated from the AKR1B10 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
