Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Biotin Anti-Melanoma gp100 antibody [EP4863(2)] (ab204674)
IHC image of Melanoma gp100 staining in a section of formalin-fixed paraffin-embedded human skin melanoma*. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20mins, performed on a Leica BONDTM. The section was then incubated overnight at +4°C with ab204674 at 1/50 dilution. Detection was via an HRP conjugated ABC system and DAB was used as the chromogen (ab103723), diluted 1/100 and incubated for 10min at room temperature. The section was then counterstained with haematoxylin and mounted with DPX.
The inset negative control image is taken from an identical assay without primary antibody.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
