Antibody Biotinylation Check Kit (ab270030) is a quick immunochromatography test that allows you to confirm the successful antibody biotinylation in one easy step, and without the need for any specialized or costly equipment. Unlike traditional methods of assessing the success of biotinylation, such as HABA assays, which can be tedious and time consuming, this kit requires only 10 minutes to perform and very small quantities of your antibody conjugate.
The key component of the kit is a nitrocellulose membrane containing a Test line of immobilized Protein A and Protein G with high affinity for the Fc region of a variety of IgG molecules, and a 40 nm Gold-Streptavidin conjugate for detection. When the biotinylated antibody is mixed with the Gold-Streptavidin conjugate, it forms a complex that flows along the nitrocellulose membrane and binds to the Protein A and Protein G on the Test line. A visible red line appears on the strip confirming successful conjugation.

How the kit works:
The nitrocellulose membrane contains a Test line of immobilized Protein A and Protein G. Both Protein A and Protein G have a high affinity for the Fc region of a variety of IgG molecules. The membrane also contains an absorbent pad to promote and control the flow of sample through the nitrocellulose. The biotinylated antibody is run on the Protein A/G Strip together with 40 nm gold conjugated streptavidin as the detection label. Biotin binds to streptavidin with exceptionally high affinity and when the biotinylated antibody flows along the nitrocellulose, upon binding to the Protein A and Protein G concentrated on the Test line, a visible red line appears on the strip.

Polyclonal antibodies from rabbit, goat, mouse and rat have been conjugated with Lightning-Link® Rapid Biotin A and run on Protein A / G strips at different concentrations, together with 0.4OD gold conjugated streptavidin. Antibodies from all species provided a red visible line on the strips.
