why is the actual band size different from the predicted?

Exposure time: 3 minutes

The expression profile observed is consistent with what has been described in the literature, the band of 45 KDa represents the full length Wnt16, the band of 32 KDa represents the degraded form of Wnt16. (PMID:26254351, 17200136).

Negative control: U-2 OS (PMID: 25999350).

This blot was developed using a higher sensitivity ECL substrate.

Immunoprecipitation - Anti-Wnt16 antibody [EPR22889-129] (ab269278)

Wnt16 was immunoprecipitated from 0.35 mg HEK-293 (human embryonic kidney epithelial cell) whole cell lysate using ab269278 at 1/30 dilution. Western blot was performed on the immunoprecipitate using ab269278 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used as the secondary anyibody at 1/5000 dilution.

Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab269278 in HEK-293 whole cell lysate
