Lanes 1-4 : Anti-PSD95 antibody (ab12093) at 2 µg/ml (Blocked with 2% BSA)
Lanes 5-8 : Anti-PSD95 antibody (ab12093) at 2 µg/ml (Blocked with 3% Milk)
Lanes 1 & 5 : Human brain cerebral cortex tissue lysate - total protein (ab30061)
Lanes 2 & 6 : Human brain tissue lysate - total protein
Lanes 3 & 7 : Brain (Mouse) Tissue Lysate
Lanes 4 & 8 : Brain (Rat) Tissue Lysate
Lysates/proteins at 25 µg per lane.
Secondary
All lanes : Rabbit polyclonal to Goat IgG - H&L - Pre-Adsorbed (HRP) at 1/5000 dilution
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 80 kDa
Observed band size: 95 kDa why is the actual band size different from the predicted?
Additional bands at: 110 kDa, 38 kDa, 55 kDa. We are unsure as to the identity of these extra bands.
Exposure time: 4 minutesThis blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin or 3% Milk before being incubated with ab12093 overnight at 4°C. Antibody binding was detected using an anti-goat antibody conjugated to HRP, and visualised using ECL development solution ab133406.
Abcam recommends using milk as the blocking agent. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.

ab12093 PSD95 antibody immunostaining brain tissue (4% PFA perfused paraffin embedded 5 micron saggital section). ab12093 was used at 10 µg/ml for a 20 minute incubation at 37 degrees C (on a Ventana Discovery slide processor). Light antigen retrieval and then light protease treatment is recommended before addition of primary antibody ab12093 in this paradigm.
