Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized LLC1 (mouse lung carcinoma) cells labeling p53 with ab246223 at 1/100 dilution, followed by a AlexaFluor®488 Goat anti-Rabbit secondary (ab150077, green) at a 1/1000 dilution. p53 was immunoprecipitated from 0.35 mg LLC1 (Mouse lung carcinoma) whole cell lysate with ab246223 at 1/30 dilution. Western blot was performed on the immunoprecipitate using ab246223 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/5000 dilution.
Lane 1: LLC1 whole cell lysate (Input).
Lane 2: ab246223 IP in LLC1 whole cell lysate.
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab246223 in LLC1 whole cell lysate.Blocking and dilution buffer and concentration: 5% NFDM/TBST.
Exposure time: 3 seconds.

Flow cytometric analysis of 4% paraformaldehyde-fixed, 90% methanol permeabilized, LLC1 (Mouse lung carcinoma) cell line labeling p53 with ab246223 at 1/60 dilution (Red) compared with a Rabbit monoclonal IgG (ab172730, Black) isotype control and an unlabeled control (Cell without incubation with primary antibody and secondary antibody, Blue). A Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1/2000 dilution.
