Immunogen
Synthetic peptide corresponding to Human IP3 receptor aa 2700 to the C-terminus (C terminal) conjugated to keyhole limpet haemocyanin.
(Peptide available asab107795) -
Positive control
- This antibody gave a positive signal in HeLa and PC3 Whole Cell Lysates. A band of the expected size was also detected in MALME 3M and Raji Whole Cell Lysates (data not shown). In our hands, the signal was relatively weak in these lysates.

General notes
The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.
One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Function
Intracellular channel that mediates calcium release from the endoplasmic reticulum following stimulation by inositol 1,4,5-trisphosphate. -
Tissue specificity
Widely expressed. -
Involvement in disease
Defects in ITPR1 are the cause of spinocerebellar ataxia type 15 (SCA15) (SCA15) [MIM:606658]. Spinocerebellar ataxia is a clinically and genetically heterogeneous group of cerebellar disorders. Patients show progressive incoordination of gait and often poor coordination of hands, speech and eye movements, due to degeneration of the cerebellum with variable involvement of the brainstem and spinal cord. SCA15 is an autosomal dominant cerebellar ataxia (ADCA). It is very slow progressing form with a wide range of onset, ranging from childhood to adult. Most patients remain ambulatory. -
Sequence similarities
Belongs to the InsP3 receptor family.
Contains 5 MIR domains. -
Domain
The receptor contains a calcium channel in its C-terminal extremity. Its large N-terminal cytoplasmic region has the ligand-binding site in the N-terminus and modulatory sites in the middle portion immediately upstream of the channel region. -
Post-translational
modificationsPhosphorylated by cAMP kinase. Phosphorylation prevents the ligand-induced opening of the calcium channels.
Phosphorylated on tyrosine residues. -
Cellular localization
Endoplasmic reticulum membrane. - Information by UniProt

Lane 1: Wild-type HAP1 whole cell lysate (20 µg)
Lane 2: IP3 knockout HAP1 whole cell lysate (20 µg)
Lane 3: SHSY-5Y whole cell lysate (20 µg)Lanes 1 - 3: Merged signal (red and green). Green - ab97823 observed at 314 kDa. Red - loading control, ab18058, observed at 130 kDa.
ab97823 was shown to specifically recognize IP3 receptor in wild-type HAP1 cells along with additional cross reactive bands. No band was observed when IP3 knockout cells were examined. Wild-type and IP3 receptor knockout samples were subjected to SDS-PAGE. Ab97823 and ab18058 (Mouse anti Vinculin loading control) were incubated overnight at 4°C at 1 µg/ml and 1/20,000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) secondary antibodies at 1/20,000 dilution for 1 hour at room temperature before imaging.
