ChIP analysis using HeLa cells, ab195482 and optimized PCR primer sets for qPCR. ChIP was performed using sheared chromatin from 1 million cells. A titration of the antibody consisting of 1, 2, 5, and 10 µg per ChIP experiment was analysed. IgG (2 µg/IP) was used as negative IP control. QPCR was performed with primers specific for promoter of the inactive HBB gene and the coding regions of the inactive MYOD gene, used as positive controls, and for the promoters of the active genes c-fos and GAPDH, used as negative controls.

Dot Blot analysis was performed to test the cross reactivity of ab195482 against Histone H3 (di methyl K9) (H3K9me2) with peptides containing other modifications of histone H3 and the unmodified H3K9 sequence. One hundred to 0.2 pmol of peptide containing the respective histone modification were spotted on a membrane. ab195482 was used at 1/20,000 dilution.
