Flow cytometric analysis of 4% paraformaldehyde-fixed RAW 264.7 (Mouse macrophage cell line transformed with Abelson murine leukaemia virus) cell line labeling CD11b with ab184307 at 1/120 dilution (red) compared with a Rabbit IgG,monoclonal [EPR25A]- Isotype control (ab172730; black) and an unlabelled control (cells without incubation with primary antibody and secondary antibody; (blue). Goat anti Rabbit IgG (FITC) at 1/500 dilution was used as the secondary antibody.

Flow cytometric analysis of 4% paraformaldehyde-fixed WEHI-231 (Mouse lymphoblast B cell lymphoma cell line) cells labeling CD11b with ab184307 at 1/120 dilution (red) compared with a Rabbit IgG,monoclonal [EPR25A]-Isotype control (ab172730) (black) and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat anti Rabbit IgG (FITC) at 1/500 dilution was used as the secondary antibody.
Negative control: WEHI-231 is negative for CD11b.

Lanes 1-3 : Anti-CD11b antibody [EPR19373] (ab184307) at 1/1000 dilution
Lanes 4-5 : Anti-CD11b antibody [EPR19373] (ab184307) at 1/5000 dilution
Lane 1 : Mouse spleen lysate
Lane 2 : M1 (Mouse myeloblast myeloid leukaemia cell line) whole cell lysate
Lane 3 : WEHI-231 (Mouse lymphoblast B cell lymphoma cell line) whole cell lysate
Lane 4 : RAW 264.7 (Mouse macrophage cell line transformed with Abelson murine leukemia virus) whole cell lysate
Lane 5 : J774A.1 (Mouse macrophage reticulum cell sarcoma cell line) whole cell lysate
Lysates/proteins at 20 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/100000 dilution
Predicted band size: 127 kDa
Observed band size: 170 kDa why is the actual band size different from the predicted?
Exposure time: 3 minutesBlocking/Dilution buffer: 5% NFDM/TBST.
CD11b is a 170 kD glycoprotein also known as αM integrin, Mac-1 α subunit (PMID: 10603370). It is exclusively restricted to cells of the myeloid lineages and monocytes, but not in T or B cell lines, so WEHI-231 cell lysate serves as a negative control (PMID: 2457584).
