The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.
One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Alexa Fluor® 647 Anti-ErbB2 / HER2 antibody [EP1045Y] (ab281578)
Immunohistochemical analysis of paraffin-embedded human breast carcinoma tissue labeling ErbB2 / HER2 with ab281578 at 1/50 dilution (10.0 μg/ml).
Membranous staining on human breast carcinoma. The section was then incubated overnight at +4°C in TBS containing 0.025% (v/v) Triton X-100 and 1% (w/v) BSA with ab281578 at 1/50 dilution (shown in Red). Nuclear DNA was labeled with DAPI (shown in blue). The section was then mounted using Fluoromount®. Non-specific protein-protein interactions were then blocked in TBS containing 0.025% (v/v) Triton X-100, 0.3M (w/v) glycine and 1% (w/v) BSA for 1h at room temperature. Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
Heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0).

Immunocytochemistry analysis of 4% Paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized SK-BR-3 (human mammary gland adenocarcinoma cell line) cells labeling ErbB2 / HER2 with ab281578 at 1/250 (2.0 µg/ml) dilution (Red).
Confocal image showing membranous staining in SK-BR-3 cells.
ab195887 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 488) was used to counterstain tubulin at 1/200 dilution (Green). The nuclear counterstain was DAPI (Blue).
