Product overview
AK1 Activity Assay Kit (Colorimetric/Fluorometric) (ab211095) provides a simple, sensitive and reliable method to detect AK1 activity in cell and tissue lysates, mitochondrial lysates or purified protein sample. In this assay, ATP generated by AK1 from ADP can be detected through a multi-step reaction, via the generation of an intermediate that reacts with a probe to form an end product that can be easily quantified colorimetrically (OD = 570 nm) or fluorometrically (Ex/Em = 535/587 nm).
See Adenylate Kinase Cytotoxicity Assay Kit ab228557 for a simple luminometric assay to measure adenylate kinase in cell culture media.

Notes
AK1 (AK, ADK, myokinase, EC 2.7.4.3) is a phosphotransferase that catalyzes the interconversion of two molecules of ADP to generate ATP and AMP. AK1 is an abundant enzyme involved in energy metabolism and homeostasis of cellular adenine nucleotide ratios in different intracellular compartments. Nine isoforms of AK1 have been identified in mammals, differing in molecular weight, tissue distribution, subcellular localization, substrate and phosphate donor specificity, and kinetic properties. Erythrocyte adenylate kinase deficiency is associated with hemolytic anemia. AK1 also plays an important role in post-ischemic recovery and in apoptosis.

Function
Catalyzes the reversible transfer of the terminal phosphate group between ATP and AMP. Small ubiquitous enzyme involved in energy metabolism and nucleotide synthesis that is essential for maintenance and cell growth. -
Involvement in disease
Defects in AK1 are the cause of hemolytic anemia due to adenylate kinase deficiency [MIM:612631]. -
