Product overview
Rat Fas ELISA Kit (ab245706) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Fas protein in cell culture media, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat Fas with 26.7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Function
Receptor for TNFSF6/FASLG. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both. The secreted isoforms 2 to 6 block apoptosis (in vitro). -
Tissue specificity
Isoform 1 and isoform 6 are expressed at equal levels in resting peripheral blood mononuclear cells. After activation there is an increase in isoform 1 and decrease in the levels of isoform 6. -
Involvement in disease
Defects in FAS are the cause of autoimmune lymphoproliferative syndrome type 1A (ALPS1A) [MIM:601859]; also known as Canale-Smith syndrome (CSS). ALPS is a childhood syndrome involving hemolytic anemia and thrombocytopenia with massive lymphadenopathy and splenomegaly. -
Sequence similarities
Contains 1 death domain.
Contains 3 TNFR-Cys repeats. -
Domain
Contains a death domain involved in the binding of FADD, and maybe to other cytosolic adapter proteins. -
Cellular localization
Secreted and Cell membrane. - Information by UniProt

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The Fas standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
Interpolated concentrations of native Fas in rat unstimulated thymus tissue culture supernatant (3 days) and rat spleen tissue culture stimulated with PMA (50 ng/mL) and Ionomycin (500 ng/mL) for 3 days. The concentrations of Fas were measured in duplicates, interpolated from the Fas standard curves and corrected for sample dilution. Undiluted samples are as follows: thymus tissue supernatant 50% and spleen tissue supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Fas concentration was determined to be 5,444 pg/mL in neat thymus tissue culture supernatant and 791.4 pg/mL in spleen tissue culture supernatant. Unstimulated spleen supernatant was not tested.
