Product overview
Phospho-ATF2 (T69/71) ELISA Kit (ab279737) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated ATF2 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-ATF2. An anti-pan ATF2 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and ATF2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and rabbit anti-phospho-ATF2 (T69/71) antibody is used to detect phosphorylated ATF2. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ATF2 (T69/71) bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Function
Transcriptional activator, probably constitutive, which binds to the cAMP-responsive element (CRE) (consensus: 5'-GTGACGT[AC][AG]-3'), a sequence present in many viral and cellular promoters. Interaction with JUN redirects JUN to bind to CRES preferentially over the 12-O-tetradecanoylphorbol-13-acetate response elements (TRES) as part of an ATF2/JUN complex. -
Tissue specificity
Abundant expression seen in the brain. -
Sequence similarities
Belongs to the bZIP family. ATF subfamily.
Contains 1 bZIP domain.
Contains 1 C2H2-type zinc finger. -
Post-translational
modificationsPhosphorylation of Thr-69 and Thr-71 by MAPK14 causes increased transcriptional activity. Also phosphorylated and activated by JNK. -
