    Mouse/Rat B7-H4 ELISA Kit  (ab263879) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of B7-H4 protein in tissue culture media and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse/Rat B7-H4 with 21 pg/ml sensitivity.


    SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:


            - Single-wash protocol reduces assay time to 90 minutes or less
            - High sensitivity, specificity and reproducibility from superior antibodies
            - Fully validated in biological samples
            - 96-wells plate breakable into 12 x 8 wells strips


    A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

  • Notes

    B7-H4 is a negative regulator of T-cell mediated immune response. Increased protein levels are associated with many forms of cancer, including ovarian, breast, lung, and prostate. In concert with Treg, B7-H4 plays a key role in suppression of tumor-associated antigen-specific T-cell immunity.  The immunogen for this kit shares 99.1% and 91% sequence identity with the rat and human homologs, respectively.

  • Function

    Negatively regulates T-cell-mediated immune response by inhibiting T-cell activation, proliferation, cytokine production and development of cytotoxicity. When expressed on the cell surface of tumor macrophages, plays an important role, together with regulatory T-cells (Treg), in the suppression of tumor-associated antigen-specific T-cell immunity. Involved in promoting epithelial cell transformation.

    Overexpressed in breast, ovarian, endometrial, renal cell (RCC) and non-small-cell lung cancers (NSCLC). Expressed on activated T- and B-cells, monocytes and dendritic cells, but not expressed in most normal tissues (at protein level). Widely expressed, including in kidney, liver, lung, ovary, placenta, spleen and testis.

      The B7-H4 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

    • Interpolated concentrations of native B7-H4 in mouse uterus tissue extract and rat uterus tissue extract based on a 1,000 µg/mL and 750 µg/mL extract load.

      The concentrations of B7-H4 were measured in duplicate and interpolated from the B7-H4 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean B7-H4 concentration was determined to be 899.88 mg/mL in mouse uterus tissue extract and 1505.28 mg/mL in rat uterus tissue extract.
