Mouse XIAP ELISA Kit (ab254504) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of XIAP protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse XIAP with 32 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
XIAP plays a key role in various cellular processes, such as the regulation of cell proliferation, apoptosis, inflammation, proteasomal regulation, and immunity. One of XIAP’s predominant roles is the blocking of apoptosis through binding and direct inhibition of caspases. Increased expression of XIAP is a hallmark of some cancers including those of the lung and prostate.

Apoptotic suppressor. Has E3 ubiquitin-protein ligase activity. Mediates the proteasomal degradation of target proteins, such as caspase-3, SMAC or AIFM1. Inhibitor of caspase-3, -7 and -9. Mediates activation of MAP3K7/TAK1, leading to the activation of NF-kappa-B. -

Involvement in disease
Defects in XIAP are the cause of lymphoproliferative syndrome X-linked type 2 (XLP2) [MIM:300635]. XLP is a rare immunodeficiency characterized by extreme susceptibility to infection with Epstein-Barr virus (EBV). Symptoms include severe or fatal mononucleosis, acquired hypogammaglobulinemia, pancytopenia and malignant lymphoma. -

Domain
The first BIR domain is involved in interaction with TAB1/MAP3K7IP1 and is important for dimerization. The interactions with SMAC and PRSS25 are mediated by the second and third BIR domains. -

Interpolated concentrations of native XIAP in NIH3T3 cell extract and mouse brain extract. The concentrations of XIAP were measured in duplicate and interpolated from the XIAP standard curve.
The concentrations of XIAP were measured in duplicate and interpolated from the XIAP standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean XIAP concentration was determined to be 2.51 ng/mL in NIH3T3 cell extract, and 1.70 ng/mL in mouse brain extract.
