Abcam’s TRAIL (TNF-related apoptosis-inducing ligand) Mouse ELISA Kit (ab193730) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse TRAIL in serum, plasma and cell culture supernatants. (GeneCards: TNFRSF10D)
This assay employs an antibody specific for mouse TRAIL coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures TRAIL present in the samples. The wells are washed and biotinylated anti-mouse TRAIL antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of TRAIL bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Relevance
TRAIL is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells, but does not appear to kill normal cells although it is expressed at a significant level in most normal tissues. TRAIL binds to several members of TNF receptor superfamily including TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly also to TNFRSF11B/OPG. The activity of TRAIL may be modulated by binding to the decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and TNFRSF11B/OPG that cannot induce apoptosis. The binding of TRAIL to its receptors has been shown to trigger the activation of MAPK8/JNK, caspase 8, and caspase 3. -
